Dr Muthineni Rajini completed her MBBS from Kakatiya Medical College, Warangal, in 2003 and a DGO (Diploma in Obstetrics and Gynecology) at the Government Maternity Hospital, Hanmakonda, in 2006. She further advanced her expertise by earning a DNB in Obstetrics and Gynecology from Yashoda Super Speciality Hospitals, Secunderabad, in 2011.

In 2017, Dr Rajini undertook an ICOG-certified course in Gynecological Endoscopy at Max Cure Suyosha (Medicover) Hospitals, Hyderabad, under the mentorship of Padma Shri awardee Dr Manjula Anagani.
